By reducing this protein, senescent (aging) cells regain their youthful properties, potentially slowing or even reversing the aging process.

As humans age, the accumulation of old or “senescent” cells contributes to age-related diseases. These cells stop dividing and become dysfunctional, producing inflammatory chemicals that harm surrounding tissues. However, researchers found that by reducing AP2A1 levels, senescent cells returned to a more youthful state, resuming normal function and division.

In theory, this breakthrough could lead to the removal of age-related diseases such as Alzheimer’s and arthritis by reversing cellular senescence. Professor Richard Faragher, an expert on cellular aging from the University of Brighton, explains normal cells monitor their number of divisions as a cancer prevention mechanism. Over time, cells become senescent, halting further division and releasing inflammatory molecules that degrade surrounding tissues, effectively making them toxic to the body.

One notable effect of senescence is the significant enlargement of cells, sometimes growing up to six times their original size. These enlarged cells develop thick, scaffold-like “stress fibers” that provide additional support but also contribute to aging. Scientists used RNA interference to inhibit AP2A1 production, leading to a reduction in cell size, rejuvenation, and restored division. Conversely, increasing AP2A1 levels accelerated the aging process in young cells.
